What Is Guanfacine ER?
Guanfacine ER is an extended-release prescription medication containing guanfacine hydrochloride. It belongs to a class of medicines known as central alpha-2A adrenergic receptor agonists and is used in the treatment of Attention Deficit Hyperactivity Disorder (ADHD).
Unlike stimulant ADHD medications such as methylphenidate and amphetamine products, guanfacine ER is a non-stimulant treatment option. It may be prescribed on its own or alongside a stimulant medication when clinically appropriate.
How Does Guanfacine ER Work?
It acts on alpha-2A adrenergic receptors in the central nervous system. Its effects on these receptors are thought to contribute to improvements in attention and impulse-control symptoms associated with ADHD.
The extended-release formulation is designed for once-daily oral administration. The tablet should be swallowed whole and should not be crushed, chewed or broken because doing so can alter the rate at which guanfacine is released.

Important Safety Information
It can cause low blood pressure, slow heart rate and fainting. Sedation and tiredness can also occur. Patients taking guanfacine ER may require monitoring of blood pressure and heart rate, particularly during dose adjustments.
This medication also interacts with certain medicines that affect CYP3A4. Strong or moderate CYP3A4 inhibitors can increase its exposure, while CYP3A4 inducers can reduce exposure.
